不同萱草属植物花瓣罗勒烯释放规律及基因表达分析

Analysis of the release of ocimene from petals and the expression of ocimene synthetase gene in different species of Hemerocallis spp.

  • 摘要: 探究罗勒烯合成酶基因参与调控罗勒烯挥发物生物合成的分子机制,为培育萱草和黄花菜的芳香型品种提供理论参考。采用顶空固相微萃取(HS-SPME)结合气相色谱—质谱联用(GC–MS)技术和分子生物学手段,分析萱草属2个萱草品种(‘醉香妃’‘香草’)、2个黄花菜品种(‘武平早’‘丁庄大菜’)的关键致香成分罗勒烯的释放规律以及其合成的关键基因的表达差异。萱草‘醉香妃’‘香草’罗勒烯含量在盛开期最高,分别为13.9、192.84 ng·g–1,而黄花菜‘武平早’‘丁庄大菜’在花苞期最高,分别达1222.96、280.52 ng·g–1,其含量约是萱草‘香草’的124、28倍。在500 μmol/L的茉莉酸甲酯(MeJA)溶液处理后,‘醉香妃’萱草罗勒烯达到最大释放量,显著高于对照组,是对照组的1.8倍;在15 ℃条件下,罗勒烯释放量最高,显著高于10和20 ℃。分别从‘醉香妃’‘香草’‘武平早’‘丁庄大菜’中各分离2个罗勒烯合成酶基因,分析显示,所有成员均具有DDxxD结构域,除‘丁庄大菜’HfTPS-dz1外,其余成员均含有NSE/DTE结构域;萱草和黄花菜品种的罗勒烯合成酶基因的表达水平与花瓣罗勒烯含量具有显著正相关性,其中‘丁庄大菜’HfTPS-dz1的关联性最低。研究结果表明,罗勒烯合成酶基因参与罗勒烯合成,但是其贡献度在品种之间具有一定差异。

     

    Abstract: The study aims to explore the molecular mechanism of ocimene synthase gene involved in the biosynthesis of volatile compounds of ocimene, and to provide theoretical reference for the cultivation of aromatic varieties of Hemerocailis fulva and Hemerocailis citrina. Using headspace solid phase microextraction (HS-SPME) combined with gas chromatography-mass spectrometry (GC-MS) and molecular biology methods, the release of ocimene and the expression of bocimene synthase gene in two H.fulva cultivars (‘Zuixiangfei’ and ‘Villa Vanilla’) and two H.citrina cultivars (‘Wupingzao’ and ‘Dingzhuang Dacai’) were analyzed. The results showed that the ocimene content of H.fulva (‘Zuixiangfei’ and ‘Villa Vanilla’) was the highest in blooming stage, reaching 13.9 ng·g–1 and 692.84 ng·g–1, respectively, while the content of H.citrina (‘Wupingzao’ and ‘Dingzhuang Dacai’) was the highest in bud stage, reaching 1222.96 ng·g–1 and 280.52 ng·g–1, respectively, and their contents were about 124 and 28 times that of H.fulva (‘Villa Vanilla’). After treatment with 500 μmol/L methyl jasmonate (MeJA) solution, the maximum release of H.fulva (‘Zuixiangfei’) ocimene was significantly higher than that of the control group, which was 1.8 times the amount of the control group. The release of ocimene was the highest at 15 ℃, significantly higher than that at 10 ℃ and 20 ℃.Two ocimene synthetase genes were isolated from ‘Zuixiangfei’, ‘Villa Vanilla’, ‘Wupingzao’, and ‘Dingzhuang Daicai’ respectively. Analysis revealed that all members possessed the DDxxD domain, and all except HfTPS-dz1 from 'Dingzhuangdacai' contained the NSE/DTE domain. There was a significant positive correlation between the expression level of ocimene synthase gene and the content of ocimene in petals of Hemerocailis fulva and Hemerocailis citrina, among which the correlation between ‘Dingzhuang Dacai’ HfTPS-dz1 was the lowest. Research results showed that the ocimene synthase gene was involved in the synthesis of ocimene, but its contribution was different among the cultivars.
