不同萱草品种的染色体数目鉴定及核型分析

Identification of chromosome numbers and karyotype analysis of Hemerocallis

  • 摘要: 以30个萱草品种为试材,采用染色体压片法对其进行染色体数目鉴定,并选择其中8个不同株型和花色的品种进行核型特征分析和聚类分析,以期为萱草品种分类、鉴定及育种实践提供细胞学依据。结果表明,30个萱草品种中有20个二倍体(2n = 2x = 22)和10个四倍体(2n = 4x = 44),染色体基数为x = 11。‘摩洛哥日出’和‘热带惊喜’是四倍体,与美国萱草协会登记的倍性不符;其余品种的倍性与先前报道一致。8个品种的核型均为“2B”型,核不对称系数范围为59.58% ~ 65.35%,均未观察到随体的存在。在8个品种中,‘波久’的核型不对称系数最大,进化程度较高;‘印度天空’的核型不对称系数最小,进化程度较低。聚类结果显示,遗传距离为5时,可将8个萱草品种分为3类。其中,第Ⅲ类仅有‘波久’这一个品种,该品种与其他7个品种亲缘关系最远。

     

    Abstract: The chromosome numbers of 30 Hemerocallis cultivars were observed and determined by using the conventional squash method. Among them, 8 varieties with different plant types and flower colors were chosen for chromosomes karyotype analysis and cluster analysis, aiming to provide a cytological basis for the classification, identification, and breeding practices of Hemerocallis cultivars. The results showed that there were 20 diploids (2n = 2x = 22) and 10 tetraploids (2n = 4x = 44) among the 30 Hemerocallis cultivars, with the chromosome base of x = 11. ‘Moroccan Sunrise’ and ‘Tropical Surprise’ were tetraploids, which differed from the ploidy registered in the American Daylily Association. The chromosome Numbers of other 28 cultivars were consistent with previous reports. The karyotype types of the 8 cultivars were all "2B" type. The asymmetrical karyotype coefficients of 8 cultivars ranged from 59.58% to 65.35%, none satellite was observed in the metaphase nuclei of these resources. Among the 8 cultivars, ‘Pojo’ had the highest degree of karyotype evolution, while ‘Indian Sky’ had the lowest. The clustering results showed that when the genetic distance was 5, the 8 Hemerocallis cultivars could be divided into 3 categories. The third category only had one cultivar ‘Pojo’, which had the farthest genetic relationship with the other 7 cultivars.
