Matrix-Assisted Laser Desorption Ionization Mass Spectrometry Imaging and Its Application

  • 摘要: 质谱成像技术是在质谱技术的基础上发展起来的新兴研究领域,其将质谱技术与计算机成像软件相结合,能使被测物质分子可视化。由于该技术无需特殊染色、对比标记、复杂的样品前处理就可以在原位观察分子的空间分布的优点,在植物组织结构研究、药物代谢、临床诊断、司法检测等领域表现出良好的应用前景。综述了基质辅助激光解吸电离质谱成像(MALDI-MSI)的原理、操作流程、样品处理、基质选择与喷涂,以及近年来在食品分析、药物分析、医学领域、植物研究中的应用。

     

    Abstract: Mass spectrometry imaging technology is an emerging research field developed on the basis of mass spectrometry technology. It combines mass spectrometry technology with imaging software technology, which can visualize the molecules of the measured substances. This technique can be used to observe the spatial distribution of molecules in situ without special staining, contrast labeling and complex sample pretreatment. It has a good application prospect in the fields of plant tissue structure research, drug metabolism, clinical diagnosis, judicial detection and so on. The principle, operation process, sample processing, matrix selection and spraying of matrix-assisted laser desorption ionization mass spectrometry imaging were reviewed, and its applications in food analysis, pharmaceutical analysis, medical field and plant research in recent years were summarized as well.
