细叶石斛组培快繁技术的优化

Optimization of tissue culture and rapid propagation technology of Dendrobium hancockii Rolfe

  • 摘要: 以细叶石斛(Dendrobium hancockii Rolfe)成熟种子为外植体,研究不同激素组合对原球茎途径的增殖、分化以及壮苗生根的影响。结果表明:细叶石斛种子萌发培养50 d即可出现大量饱满绿色的原球茎;增殖、分化阶段最佳培养基为MS培养基+1.0 mg·L–1 6-苄氨基嘌呤(6-BA)+0.3 mg·L–1 萘乙酸(NAA)+蔗糖30 g·L–1+琼脂7.8 g·L–1,增殖率为16.67%;生根壮苗最适培养基为MS培养基+0.5 mg·L–1 NAA+0.1 mg·L–13吲哚丁酸(indolebutyric acid,IBA),生根率100%,分化系数为6.14,幼苗健壮且生长快速。优化了细叶石斛组培快繁体系,为后续进一步优化以及工厂化生产提供依据。

     

    Abstract: Utilizing the mature seeds of Dendrobium hancockii Rolfe as explants, this study investigated the impacts of various hormone combinations on the proliferation, differentiation, and rooting of protocorm-like bodies (PLBs). The results indicated that a large number of plump and green PLBs emerged from the Dendrobium hancockii Rolfe seeds after 50 days of germination cultivation. The optimal medium for proliferation and differentiation was MS medium supplemented with +1.0 mg·L-1 6-BA+0.3 mg·L-1 NAA+30 g·L-1 sucrose+7.8 g·L-1 agar, with a proliferation rate of 16.67%. The optimal medium for rooting and seedling growth was MS medium + 0.5 mg·L-1 NAA+0.1 mg·L-1IBA, with a rooting rate of 100% and a differentiation coefficient of 6.14, resulting in robust seedlings with rapid growth. This study optimized the tissue culture and rapid propagation system of Dendrobium hancockii Rolfe, providing a basis for further optimization and industrial production.
