香果树愈伤组织诱导及抗褐化研究

Study on Embryogenic Callus Induction and Anti-Browning of Emmenopterys henryi Oliv.

  • 摘要: 以珍稀濒危植物香果树的叶片、叶柄、茎段为外植体,进行胚性愈伤组织诱导,并针对愈伤组织继代培养中褐化严重的问题进行抗褐化剂的筛选。实验结果表明,叶柄是香果树愈伤组织诱导的最适外植体,依次用75%酒精处理60 s、0.2% HgCl2处理15 min灭菌效果最好,并选定了香果树愈伤组织最适诱导培养基和继代培养基。

     

    Abstract: The leaf, petiole and stem segments of Emmenopterys henryi Oliv. were used for the induction of embryogenic callus. To alleviate callus browning in subculture of callus, anti-browning agents were screened. Results showed that petioles sterilized by 75% alcohol for 60 s and 0.2% HgCl2 for 15 min successively were optimal for embryonic callus induction. The optimum formula for embryogenic callus induction and callus browning inhibition during subculture were decided.
